What is Inflammation?

The body’s natural immune response, defending us against foreign invaders by fighting off infection and illness. Although inflammation is a normal and necessary process, it can become chronic and lead to long-term health issues if left unresolved.

What are the Phases of Inflammation?

  1. Initiation Phase: Tissue is negatively affected (such as over-exercising muscle), initiating the body’s natural protective response.

· Eicosanoid lipid mediators (i.e. LTB4, PGE2), cytokines, chemokines, and other

messengers are released to increase the inflammatory and immune response

· White blood cells (such as neutrophils) enter the affected area and mount the

body’s response

· Redness or swelling may occur

2. Resolution Phase: Specialized Pro-Resolving Mediators (SPMs) are produced within

tissue by immune cells and function as “resolution agonists” to promote resolution

(i.e. completion) of inflammation and physical challenges.

· SPMs reduce excessive white blood cells activity

· SPMs trigger macrophages to remove dead neutrophils, bacteria, and debris

· SPMs aid in tissue remodeling

· SPMs support return to homeostasis and normal immune surveillance

· Immune cells, which help clean-up the affected area, can be enhanced by SPMs

· When the body is unable to complete the resolution phase, chronic

inflammation occurs

What causes Chronic Inflammation?

· Stress

· Food Allergens

· Environmental Irritants and Toxins

· Lack of Exercise and Physical Fitness

· Physical Stressors

· Lifestyle Behaviors

· Poor Diet and Nutrition

· Dietary Patterns

· Aging Process

What are Specialized Pro-Resolving Mediators (SPMs)?

· SPMs are produced naturally in the body from unsaturated fatty acids,

predominantly omega-3 EPA and DHA

· EPA and DHA are precursors to SPMs- they require multiple enzymatic

conversions to form SPMs and have pro-resolving properties

· Proprietary extract from fish oil that isolates the SPMs 17-HDHA and 18-HEPE

found in marine lipids at a standard concentration

· Specialized Pro-Resolving Mediators found in OmegaGenics SPM Active are a

group of lipid mediators that function as “resolution agonists” by actively helping

facilitate the body’s natural resolution process and completion of its response to

physical challenges

· SPMs promote healing and help the body’s inflammation return to homeostasis

SPMs Mechanism of Action

During the resolution phase, SPMs are produced at the affected tissue site and function as “resolution agonists,” by orchestrating the resolution-related activity and helping to facilitate this response toward homeostasis by:

· Limiting excessive white blood cells infiltration in affected tissue

· Stimulating the clean up of cellular debris

· Decreasing pro-inflammatory mediator production

· Enhancing tissue regeneration and restoring to homeostasis

Role of SPMs in the Body

· Reprogram immune cells: SPMs encourage immune cells to be more protective and

less likely to amplify the negative effects of an environmental stressor

· Help restore balance: SPMs are necessary to return the body to homeostasis after a

physical challenge and support overall wellbeing

· Support resilience: SPM resolution activities play a role in the body’s ability to bounce

back from environmental challenges

Potential Clinical Applications

· Osteoarthritis: Characterized by an increase in inflammatory cells and biomarkers in

affected joints

· Cognition: Neuroinflammation has been associated with cognitive decline

· Metabolic Disease: Fat tissue dysfunction contributes to chronic uncontrolled

inflammation, which can result in adverse metabolic outcomes such as insulin

resistance and obesity

· Inflammatory Bowel Disorders: Crohn’s disease and ulcerative colitis are irritable

bowel diseases that lead to long-term and occasionally irreversible impairment of gut

structure and function

SPMs Dosage

Severe Conditions:

· Take 6 soft gels daily for 1 week, then take 4 soft gels daily for 1 week, then take

2 soft gels daily

Mild to Moderate Conditions:

· Take 2 soft gels daily

Onset of Action

· Healthy individuals had an increase in circulating SPMs within 2 hours

· In healthy individuals, 24 hours after SPM Active consumption the behavior of their

immune cells shifted to a more protective response as demonstrated by an increase in

clean up of bacteria

Why Metagenics SPM Active?

· Proprietary blend of SPMs that average 300% bioactivity compared to other

competitors

· SPM Active bioavailability has been confirmed in multiple studies

· SPM Active helps reprogram the immune cells so that they are less likely to amplify the

negative effects of an environmental stressor/trigger, and more likely to be protective

· In healthy individuals within 24 hours of consuming SPM Active, the immune cells

shifted their behavior to be more protective and less damaging, which represents a

more resilient response

· SPMs’ resolution activities are critical processes of the body’s natural healing capacity

and innate intelligence and resilience, helping to recover from physical and

environmental challenges

· Immune cells of individuals with excess fat tissue have been shown to have reduced

ability to produce resolvins from DHA. Providing these cells with17-HDHA increases

resolvin production for a healthier response and helps restore the balance between markers of immune response initiation and markers of resolution in the cells

· Research in healthy human immune cells shows that 17-HDHA and 18-HEPE are more

efficient at promoting resolving production that EPA and DHA

· By targeting critical resolution activities, supplementation with SPM Active is part of a

systems based approach to promote good health

· Supplementation with SPM-Active led to higher levels of circulating SPMs
